Systemic characteristics of biomarkers and differential metabolites of raw and ripened pu-erh teas by chemical methods combined with a UPLC-QQQ-MS-based metabolomic approach

Pu-erh tea is one of the most popular beverages in China and Southeast Asia, however, influences from fermentation and storage on its chemical profile and quality are unclear. Thus, bioactivities and metabolomes of raw (17-raw) and ripened teas (17-rip through 06-rip) were assessed using chemical methods and a UPLC-QQQ-MS-based metabolomic approach. Results evidence that chemical components and antioxidant activities of 17-rip through 06-rip were similar but lower than those of 17-raw. Subsequently, 842 metabolites were identified from 17-raw, 17-rip and 06-rip, of which 20 and 19 metabolites were biomarkers for discerning 17-rip from 17-raw and 06-rip, respectively. Between 17-raw and 17-rip, 536 differential metabolites were identified, and 17-rip contained higher levels of gallic acid, acetyl amino acids, purine alkaloids, pyrimidine alkaloids and non glycoside flavonoids and lower levels of sour compounds, quinic acid derivatives, amino acids, flavonoids glycosides, and flavan-3-ols; all of them were found to be positively but gallic acid and acetyl amino acids negatively correlated with two tested bioactivities. Between 17-rip and 06-rip, 175 differential metabolites were identified, among which alkaloids positively correlated to the two bioactivities. Overall, this study identified biomarkers distinguishing teas with different fermentation processes and storage times and different metabolites affecting their tastes and bioactivities.

Environmental chemicals and metabolic disruption in primary and secondary human parathyroid tumors

Background: The incidence of primary hyperparathyroidism has increased 300% in the United States in the past 30 years, and secondary hyperparathyroidism is almost universal in patients with end-stage renal disease. We assessed the presence of environmental chemicals in human hyperplastic parathyroid tumors as possible contributing factors to this increase. Methods: Cryopreserved hyperplastic parathyroid tumors and normal human parathyroids were analyzed by gas chromatography and liquid chromatography coupled to ultra-high-resolution mass spectrometry, bioinformatics, and biostatistics. Results: Detected environmental chemicals included polychlorinated biphenyls, polybrominated diphenyl ethers, dichloro-diphenyl-trichloroethane derivatives, and other insecticides. A total of 99% had p,p’-dichlorodiphenyldichloroethylene. More than 50% contained other environmental chemicals, and many classified as endocrine disruptors. Polychlorinated biphenyl-28 and polychlorinated biphenyl-49 levels correlated positively with parathyroid tumor mass. Polybrominated diphenyl ether-47 concentrations in tumors were inversely correlated with patients’ serum calcium levels. Cellular metabolites in pathways of purine and pyrimidine synthesis and mitochondrial energy production were associated with tumor growth and with p,p’-dichlorodiphenyldichloroethylene in primary hyperparathyroidism tumors. In normal parathyroids, p,p’-dichlorodiphenyldichloroethylene, polychlorinated biphenyl-28, polychlorinated biphenyl-74, and polychlorinated biphenyl-153, but not p,p’-dichlorodiphenyldichloroethylene or polychlorinated biphenyl-49, were detected. Conclusion: Environmental chemicals are present in human parathyroid tumors and warrant detailed epidemiologic and mechanistic studies to test for causal links to the growth of human parathyroid tumors. Conidial UVB resistance decreased by 54% in Delta wc1 and 67% in Delta wc2. Five-hour exposure of UVB-inactivated conidia to visible light resulted in photoreactivation rates of 30% and 9% for the Delta wc1 and Delta wc2 mutants, contrasting to 79%-82% for wild-type and complemented strains. Importantly, abolished transcription of phr1 in Delta wc-2 and of phr2 in Delta wc1 resulted in incapable photorepair of CDP and 6-4PP DNA lesions in UVB-impaired Delta wc2 and Delta wc1 cells respectively. Yeast two-hybrid assays revealed interactions of either WC protein with both PHR1 and PHR2. Therefore, the essential roles for WC1 and WC2 in both photorepair of UVB-induced DNA lesions and photoreactivation of UVB-inactivated conidia rely upon their interactions with, and hence transcriptional activation of, PHR1 and PHR2. These findings uncover a novel WC-cored pathway that mediates filamentous fungal response and adaptation to solar UV irradiation.

In silico design novel dihydropyrimio[4, 5-d]pyrimidine derivatives as inhibitors for colony-stimulating factor-1 receptor based on 3D-QSAR, molecular docking and molecular dynamics simulation

Colony-stimulating factor-1 receptor (CSF-1R) can affect the further development of tumors by regulating the generation and biological activity of mononuclear-phagocytic lineage cells, and is an important potential target for cancer treatment. In this study, we selected 48 dihydropyrimio [4, 5-d]pyrimidine derivatives and performed 3D-QSAR studies on them using CoMFA and CoMSIA model. The results showed that CoMFA (n = 10; q(2) = 0.626; r(2) = 0.998) and CoMSIA (n = 10; q(2) = 0.684; r(2) = 0.997) had good stability and predictability. The relationship between the activity and structure of the compounds was analyzed by counter maps of the steric, electrostatic and hydrophobic fields. Subsequently, molecular docking was used to explore key amino acids and docking modes at the active site. Based on these results, we designed 9 new dihydropyrimio [4, 5-d] pyrimidine derivatives and predicted their activities. The results indicated that these compounds had good predictive activities and ADME/T properties. MD simulation analysis confirmed that the residues Leu588 and Cys666 in the active site played a key role; at the same time, these compounds mainly bind to CSF-1R through hydrophobic interactions and hydrogen bonding. ASARONE-DERIVED PHENYLPROPANOIDS AND ISOQUINOLINE-DERIVED ALKALOIDS FROM THE BARK OF Duguetia pycnastera (Annonaceae) AND THEIR CYTOTOXICITIES

The phytochemical investigation of the hexane and methanol extracts from the bark of Duguetia pycnastera Sandwith (Annonaceae) afforded seven known compounds, two asarone-derived phenylpropanoids and five isoquinoline-derived alkaloids. The asarones, gamma-asarone (1-allyl-2,4,5-trimethoxybenzene) and 2,4,5-trimethoxy-styrene were isolated of the hexane extract while the aporphine alkaloids, O-methylmoschatoline, lysicamine, nornuciferidine, and guatterine N-oxide, and the benzyltetrahydroisoquinoline alkaloid, (S)-reticuline were isolated of the alkaloid fraction of the methanol extract. This is the first report of these compounds in D. pycnastera. gamma-Asarone is being reported for the first time in the Annonaceae. Nornuciferidine is described for the second time in the Annonaceae while guatterine N-oxide is the third register. The structures of the isolated compounds were established by extensive analyses using 1D and 2D NMR spectroscopy in combination with MS. The cytotoxic activity of the isolated compounds (except for nornuciferidine) was evaluated against cancer and non-cancerous cell lines, in which lysicamine was the most active compound, mainly against HL-60, HepG2, and K562 with IC50 values of 24.40, 28.86 and 38.75 mu mol L-1, respectively.

5-Nitro-2,4-Dichloropyrimidine as an Universal Model for Low-Energy Electron Processes Relevant for Radiosensitization

We report experimental results of low-energy electron interactions with 5-nitro-2,4-dichloropyrimidine isolated in the gas phase and hydrated in a cluster environment. The molecule exhibits a very rare combination of many so far hypothesized low-energy electron induced mechanisms, which may be responsible for synergism in concurrent chemo-radiation therapy of cancer. In contrast to many previous efforts to design an ideal radiosensitizer based on one mode of action, the present model molecule presents an alternative approach, where several modes of action are combined. With respect to the processes induced by the low-energy electrons, this is not a trivial task because of strong bond specificity of the dissociative electron attachment reaction, as it is discussed in the present paper. Unfortunately, low solubility and high toxicity of the molecule, as obtained from preliminary MTT assay tests, do not enable further studies of its activity in real biological systems but it can advantageously serve as a model or a base for rational design of radiosensitizers.

Mitochondrial DNA Affects the Expression of Nuclear Genes Involved in Immune and Stress Responses in a Breast Cancer Model

Tumor cells without mitochondrial (mt) DNA (rho(0) cells) are auxotrophic for uridine, and their growth is supported by pyruvate. While ATP synthesis in rho(0) cells relies on glycolysis, they fail to form tumors unless they acquire mitochondria from stromal cells. Mitochondrial acquisition restores respiration that is essential for de novo pyrimidine biosynthesis and for mitochondrial ATP production. The physiological processes that underpin intercellular mitochondrial transfer to tumor cells lacking mtDNA and the metabolic remodeling and restored tumorigenic properties of cells that acquire mitochondria are not well understood. Here, we investigated the changes in mitochondrial and nuclear gene expression that accompany mtDNA deletion and acquisition in metastatic murine 4T1 breast cancer cells. Loss of mitochondrial gene expression in 4T1 rho(0) cells was restored in cells recovered from subcutaneous tumors that grew from 4T1 rho(0) cells following acquisition of mtDNA from host cells. In contrast, the expression of most nuclear genes that encode respiratory complex subunits and mitochondrial ribosomal subunits was not greatly affected by loss of mtDNA, indicating ineffective mitochondria-to-nucleus communication systems for these nuclear genes. Further, analysis of nuclear genes whose expression was compromised in 4T1 rho(0) cells showed that immune- and stress-related genes were the most highly differentially expressed, representing over 70% of those with greater than 16-fold higher expression in 4T1 compared with 4T1 rho(0) cells. The monocyte recruiting chemokine, Ccl2, and Psmb8, a subunit of the immunoproteasome that generates MHCI-binding peptides, were the most highly differentially expressed. Early monocyte/macrophage recruitment into the tumor mass was compromised in 4T1 rho(0) cells but recovered before mtDNA could be detected. Taken together, our results show that mitochondrial acquisition by tumor cells without mtDNA results in bioenergetic remodeling and re-expression of genes involved in immune function and stress adaptation.
