The increased glutamine requirement for immune cell functions occurs concomitantly with the high glutamine utilization by host cells in virus-infected patients. The development of antiviral drugs that target glutamine metabolism must then be specifically directed at virus-infected host cells to avoid negative effects on immune functions. Therefore, the aim of this review was to describe the landscape of cellular glutamine metabolism to search for potential candidates to inhibit glutamine transport or glutamine metabolism.

Effect of homonuclear boron bonds in the adsorption of DNA nucleobases on boron nitride nanosheets

Quantum-mechanics calculations were carried out within the density functional theory (DFT) scheme, to analyze the sorbate-sorbent interaction between DNA nucleobases (purines: guanine and adenine; pyrimidines: cytosine and thymine) and hexagonal boron nitride nanosheets (pristine and non-stoichiometric) in two phases: i) gas and ii) aqueous. The resulting molecular simulations for the pristine nanosheet indicate that the four molecular sorbates prefer to be oriented perpendicular and/or parallel respect to sorbent. This geometrical effect generates adsorption energies associated to non-covalent interactions (physisorption), being the preferential adsorption sites those of N atoms of the nanosheet. According to the calculated quantum descriptors, they exhibit low chemical reactivity and work function, as well as high polarity and semiconductor-like behavior. However, the homonuclear boron bonds in the nanosheet (negatively charged) induce a strong interaction, almost three times larger than pristine nanosheet in gas phase; except for cytosine, due to this is weakly adsorbed in both phases. Moreover, the chemical reactivity and work function are reduced, whereas its conductivity (energy LHgap) and polarity were increased, since the preferential interaction site is corresponding to a B atom. Since the magnetic behavior (1.0 bohr magneton) of BN nanosheet/rB is not altered, the nanosheets with homonuclear boron bonds might be used as potential drug delivery vehicles and sensors. The surface temperature and combustion mechanism of TNDA were established through thermocouple studies. Functional characterization of the HMP-P synthase of Legionella pneumophila (Lpg1565)

A high density and insensitive fused [1,2,3]triazolo-pyrimidine energetic material

The copper(II) complexes of new anthrahydrazone ligands: In vitro and in vivo antitumor activity and structure-activity relationship

Two new copper(II) complexes, 9-PMAH-Cu (1) and 9-FPMAH-Cu (2), of anthrahydrazone were synthesized and structurally characterized, in which 9-FPMAH (9-(4′-trifluoromethyl)-pyrimidine anthrahydrazone) is the 4′-CF3 derivative of 9-PMAH (9-pyrimidine anthrahydrazone). Both complexes 1 and 2 showed similar intercalative binding modes towards DNA and might compete with the typical DNA intercalator, GelRed, in the same binding site. They could also act as topoisomerase (type I) suppressor to effectively inhibit its activity, in which complex 1 was more effective than 2. The in vitro antitumor screening indicated that complex 1 displayed much higher antiproliferative ability than 2 and cisplatin towards all the tested tumor cell lines. On the other hand, complex 1 also showed high cytotoxicity against human normal liver cell line HL-7702, suggesting it is a potential high cytotoxic antitumor candidate. While it was also suggested that the loss of activity of complex 2 might be due to the presence of 4′-CF3 on the pyrimidine ring. Studies on the cellular level showed that complex 1 could arrest the cell cycle of the most sensitive T-24 cells at G2/M phase and induced cell apoptosis. Complex 1 further showed a significant suppression on the tumor growth on the T-24 tumor xenograft mouse model, but not reduced the body weight. Especially, complex 1 could retain its coordination state in H2O even in the presence of HSA. The results suggests that complex 1 is of enough safety to be considered as a promising anticancer candidate by combining the bioactive Cu(II) and the anthrahydrazone pharmacophore.

Synthesis and antiproliferative assessments of new derivatives of isothiazolo[3,4-d]pyrimidine

Various derivatives of 5-aryl-4-imino-3-(phenylamino)-4,5-dihydroisothiazolo[3,4-d]pyrimidines (3a-f) were synthesized. The synthesis has been done through treatment of 3-amino-4-cyano-5-(phenylamino)isothiazole with various aryl isothiocyanates. The isothiazole skeleton was obtained by the reaction of malononitrile and phenyl isothiocyanate followed by chloramine treatment. Some of the synthesized dihydroisothiazolo[3,4-d]pyrimidines were tested against different cancer cell lines, including ACHN, HeLa, HL-60, MCF-7, and PC3. Malignant cells were cultured in RPMI medium and incubated with different concentrations of the mentioned compounds. Cell viability was assessed using the MTS assay. The cytotoxicities of the synthesized compounds are not significant and are probably safe for other biological use.

This review examines current trends in the use of readily accessible acyclic enaminodiones in the synthesis of heterocyclic structures, including medicinal and natural compounds. Enaminodiones are latent tricarbonyl compounds, therefore their synthetic use exploits mainly their electrophilic properties. In addition, they can act as C-nucleophiles and participate in formal (4+2) cycloaddition reactions as ambiphilic reagents, as well as key intermediates in intramolecular cyclizations. The review analyzes the literature published in 2012-2019; the systematization is based on the structure of the formed heterocycle. The bibliography of the review includes 97 sources.
