Experimental/computational assessments of steel in HCl medium containing aminocarbonitrile-incorporated polyhydroxy-functionalized pyrido[2,3-d]pyrimidine as a green corrosion inhibitor

This research deals with insights into the inhibitory effects of a new synthetic bio/green corrosion inhibitor based on aminocarbonitrile-incorporated polyhydroxy-functionalized pyrido[2,3-d]pyrimidine derivative (PPAC) on steel substrate exposed to 1 M HCl cleaning electrolyte. The electrochemical examinations resulted in mixed corrosion inhibition activity having a paramount cathodic hindrance with ca. 91% reduction in corrosion current density (i(corr)) and also 92.5% inhibition efficiency (eta%) achievement after 6 h steel exposing to 0.5 mM PPAC/ HCl solution. A new-route, in-situ multi-step image processing method was successfully created to extract the adsorbed hydrogen bubbles on the electrolyte/steel interface by developing a mathematical exponentiated model. The model based on image-based shape descriptors precisely reproduced the electrochemical data with errors of less than 13%. A low corrosion rate upon PPAC adsorption led to a considerable diminution of bubbles number, T, while their surface area, SA, increased remarkably. Agriculture organic wastes fermentation CO2 enrichment in greenhouse and the fermentation residues improve growth, yield and fruit quality in tomato

The concern about fossil fuel shortage and environmental pollution highlighted the necessity to improve the output performance in facility agriculture. We investigated the influence of a climate-smart agricultural production system, namely agriculture organic waste fermentation CO2 enrichment technology (AOWF + CO2) on greenhouse tomato production and the effect of fermentation residues from AOWF + CO2. AOWF + CO2 exhibited positive effect on tomato growth, yield and fruit quality. Soluble sugar, lycopene and carotenoid contents in tomato fruits increased by 24.1%, 53% and 129%, respectively, while the nitrate content decreased by 10.7% when compared with control. The metabolome and proteome studies revealed significant up-regulation of secondary metabolisms, and the differentially expressed proteins (DEPs) were significantly enriched in purine, pyrimidine and linoleic acid metabolism pathways. The fermentation residues also significantly increased tomato growth, yield and fruit quality. It was estimated that each hectare greenhouse furnished with AOWF + CO2 could at least reduce 17951.5 kg CO(2)eq. emission, and store 5314.5 kg CO(2)eq. in soil. In conclusion, AOWF + CO2 and its residues recycling could accelerate tomato growth and productivity. Mitochondrial DNA Affects the Expression of Nuclear Genes Involved in Immune and Stress Responses in a Breast Cancer Model

Tumor cells without mitochondrial (mt) DNA (rho(0) cells) are auxotrophic for uridine, and their growth is supported by pyruvate. While ATP synthesis in rho(0) cells relies on glycolysis, they fail to form tumors unless they acquire mitochondria from stromal cells. Mitochondrial acquisition restores respiration that is essential for de novo pyrimidine biosynthesis and for mitochondrial ATP production. The physiological processes that underpin intercellular mitochondrial transfer to tumor cells lacking mtDNA and the metabolic remodeling and restored tumorigenic properties of cells that acquire mitochondria are not well understood. Here, we investigated the changes in mitochondrial and nuclear gene expression that accompany mtDNA deletion and acquisition in metastatic murine 4T1 breast cancer cells. Loss of mitochondrial gene expression in 4T1 rho(0) cells was restored in cells recovered from subcutaneous tumors that grew from 4T1 rho(0) cells following acquisition of mtDNA from host cells. In contrast, the expression of most nuclear genes that encode respiratory complex subunits and mitochondrial ribosomal subunits was not greatly affected by loss of mtDNA, indicating ineffective mitochondria-to-nucleus communication systems for these nuclear genes. Further, analysis of nuclear genes whose expression was compromised in 4T1 rho(0) cells showed that immune- and stress-related genes were the most highly differentially expressed, representing over 70% of those with greater than 16-fold higher expression in 4T1 compared with 4T1 rho(0) cells. The monocyte recruiting chemokine, Ccl2, and Psmb8, a subunit of the immunoproteasome that generates MHCI-binding peptides, were the most highly differentially expressed. Early monocyte/macrophage recruitment into the tumor mass was compromised in 4T1 rho(0) cells but recovered before mtDNA could be detected. Taken together, our results show that mitochondrial acquisition by tumor cells without mtDNA results in bioenergetic remodeling and re-expression of genes involved in immune function and stress adaptation.

Anatomy of Base Pairing in DNA by Interacting Quantum Atoms

The formation of purine and pyrimidine base pairs (BPs), which contributes to shaping of the canonical and noncanonical 3D structures of nucleic acids, is one the most investigated phenomena in chemistry and life sciences. In this contribution, the anatomy of the bond energy (BDE) of the base-pairing interaction in 39 different arrangements found experimentally or predicted for DNA structures containing the four common nucleobases (A, C, G, T) in their neutral or protonated forms is described in light of the theory of interacting quantum atoms within the context of the quantum theory of atoms in molecules. The interplay of individual energy components involved in the three stages of the bond formation process (structural deformation, electron-density promotion, and intermolecular interaction) is studied. We recognized that for the neutral BPs, variations in the kinetic and electrostatic contributions to the BDE are rather negligible, leaving the exchange-correlation energy as the main stabilizing component. It is shown that the contribution of the exchange-correlation term can be recovered by including atoms that are formally assumed to be hydrogen bonded (primary interaction). In contrast, to recover the electrostatic component of interaction, one must consider both the primary and secondary (formally nonbonded atoms) interatomic interactions. The results of our study were employed to design new types of BPs with altered bonding anatomy. We demonstrate that improving the electrostatic characteristics of the BPs does not necessarily result in greater interaction energies if weak secondary hydrogen bonding is destroyed. However, the main tuning factor for systems with conserved interacting faces (primary interactions) is the electrostatic component of the interaction energy resulting from the secondary atom-atom electrostatics.
