Synthesis and evaluation of ursolic acid-based 1,2,4-triazolo[1,5-a]pyrimidines derivatives as anti-inflammatory agents

We report the results of muon-spin spectroscopy (mu+SR) measurements on the staggered molecular spin chain [pym-Cu(NO3)(2)(H2O)(2)] (pym = pyrimidine), a material previously described using sine-Gordon field theory. Zero-field mu+SR reveals a long range magnetically ordered ground state below a transition temperature T-N = 0.23(1) K. Using longitudinal-field (LF) mu+SR we investigate the dynamic response in applied magnetic fields 0 < B < 500 mT and find evidence for ballistic spin transport. Our LF mu+SR measurements on the chiral spin chain [Cu(pym)(H2O)(4)]SiF6 center dot H2O instead demonstrate one-dimensional spin diffusion, and the distinct spin transport in these two systems suggests that additional anisotropic interactions play an important role in determining the nature of spin transport in S = 1/2 antiferromagnetic chains. Cobalt and ruthenium complexes with pyrimidine based schiff base: Synthesis, characterization, anticancer activities and electrochemotherapy efficiency

In this study, a new Schiff base ligand and its two M(II) complexes [CoCl center dot L(H2O)(2)]center dot 2H(2)O, [RuCl(p-cymene)L] were synthesized. The structural features were confirmed from their micro analytical, IR, UV-Vis., (HC)-H-1-C-13 NMR, TGA, X-ray diffraction analysis, mass spectral data and magnetic susceptibility measurements. The Co(II) and Ru(II) complexes displayed an octahedral geometry. In vitro anticancer activities of the Schiff base, Co(II) and Ru(II) complexes were evaluated on the human colon cancer cell line (Caco-2) and biocompatibility characteristics were determined in the L-929 (normal fibroblast cells) cell line by using the MIT assay. Furthermore, we examined the effectiveness of electrochemotherapy (ECT) on cytotoxic activities of these compounds in Caco-2 cancer cell line. According to the findings of the study, Co(II) and Ru(II) complexes showed considerable anticancer properties in the Caco-2 colon cancer cells; however, the ligand did not show significant anticancer activity. It was determined that the combined application of electroporation (EP)+complexes were much more effective than the application of complexes alone in the treatment of Caco-2 colon cancer cells. In a conclusion, the Co(II) and Ru(II) complexes, which showed significant anticancer activity in Caco-2 colon cancer cells, increased cytotoxicity levels by 2.07 and 2.12, respectively in their combined applications with EP. EZH2 is a potential prognostic predictor of glioma

The enhancer of zeste homologue 2 (EZH2) is a histone H3 lysine 27 methyltransferase that promotes tumorigenesis in a variety of human malignancies by altering the expression of tumour suppressor genes. To evaluate the prognostic value of EZH2 in glioma, we analysed gene expression data and corresponding clinicopathological information from the Chinese Glioma Genome Atlas, the Cancer Genome Atlas and GTEx. Increased expression of EZH2 was significantly associated with clinicopathologic characteristics and overall survival as evaluated by univariate and multivariate Cox regression. Gene Set Enrichment Analysis revealed an association of EZH2 expression with the cell cycle, DNA replication, mismatch repair, p53 signalling and pyrimidine metabolism. We constructed a nomogram for prognosis prediction with EZH2, clinicopathologic variables and significantly correlated genes. EZH2 was demonstrated to be significantly associated with several immune checkpoints and tumour-infiltrating lymphocytes. Furthermore, the ESTIMATE and Timer Database scores indicated correlation of EZH2 expression with a more immunosuppressive microenvironment for glioblastoma than for low grade glioma. Overall, our study demonstrates that expression of EZH2 is a potential prognostic molecular marker of poor survival in glioma and identifies signalling pathways and immune checkpoints regulated by EHZ2, suggesting a direction for future application of immune therapy in glioma.

A Chemical Probe for Dark Kinase STK17B Derives Its Potency and High Selectivity through a Unique P-Loop Conformation

STK17B is a member of the death-associated protein kinase family and has been genetically linked to the development of diverse diseases. However, the role of STK17B in normal and disease pathology is poorly defined. Here, we present the discovery of thieno[3,2-d] pyrimidine SGC-STK17B-1 (11s), a high-quality chemical probe for this understudied dark kinase. 11s is an ATP-competitive inhibitor that showed remarkable selectivity over other kinases including the closely related STK17A. X-ray crystallography of 11s and related thieno[3,2-d]pyrimidines bound to STK17B revealed a unique P-loop conformation characterized by a salt bridge between R41 and the carboxylic acid of the inhibitor. Molecular dynamic simulations of STK17B revealed the flexibility of the P-loop and a wide range of R41 conformations available to the apo-protein. The isomeric thieno[2,3-d]pyrimidine SGC-STK17B-1N (19g) was identified as a negative control compound. The >100-fold lower activity of 19g on STK17B was attributed to the reduced basicity of its pyrimidine N1.

Pyrimidine-thioindole inhibits gastric cancer cell proliferation via up-regulation of expression of tumor suppressor miR-145

Purpose: To investigate the effect of pyrimidine-thioindole on gastric cancer proliferation and the underlying mechanism of action. Methods: Cell viability and apoptosis were determined using MTT assay and Annexin V/P1 assay, respectively. Reverse transcription-polymerase chain reaction (RT-PCR) was used for the determination of expression levels of miR-145, while protein expression levels were assayed by western blotting. Results: Pyrimidine-thioindole treatment significantly inhibited the proliferation of AGS and SNU-5 cells (p < 0.05), but had no effect on the viability of GES-1 cells. Exposure to pyrimidine-thioindole at doses of 8 and 10 mu M significantly enhanced the apoptosis of AGS and SNU-5 cells (p < 0.05). Pyrimidine-thioindole exposure markedly increased the proportions of AGS and SNU-5 cells in G1 phase (p < 0.05). In AGS and SNU-5 cell lines, pyrimidine-thioindole exposure at doses of 8 and 10 mu M significantly upregulated the expression of miR-145, with higher enhancement of miR-145 expression in AGS cells than in SNU-5 cells. Moreover, pyrimidine-thioindole downregulated the expressions of MMP-2, MMP-9, c-Myc, p-PI3K and p-AKT in AGS and SNU-5 cells. Pyrimidine-thioindole treatment enhanced the expression of p21 in AGS and SNU-5 cells, relative to untreated cells (p < 0.05). Conclusion: These results suggest that pyrimidine-thioindole activates apoptotic signaling pathway, leading to reduction in cell proliferation and arrest of cell cycle. Design, synthesis and biological evaluation of thieno[3,2-d]pyrimidine derivatives containing aroyl hydrazone or aryl hydrazide moieties for PI3K and mTOR dual inhibition

Recently, PI3K and mTOR have been regarded as promising targets for cancer treatment. Herein, we designed and synthesized four series of novel thieno[3,2-d]pyrimidine derivatives that containing aroyl hydrazone or aryl hydrazide moieties. These derivatives act as PI3K/mTOR dual inhibitors, suggesting that they can be used as cancer therapeutic agents. All compounds were tested for anti-proliferative activity against four cancer cell lines. The structure-activity relationship (SAR) studies were conducted by varying the moieties at the C-6 and C-2 positions of the thieno[3,2-d]pyrimidine core. It indicated that aryl hydrazide at C-6 position and 2-aminopyrimidine at C-2 position are optimal fragments. Compound 18b showed the most potent in vitro activity (PI3K alpha IC50 = 0.46 nM, mTOR IC50 = 12 nM), as well as good inhibition against PC-3 (human prostate cancer), HCT-116 (human colorectal cancer), A549 (human lung adenocarcinoma) and MDA-MB-231 (human breast cancer) cell lines. Furthermore, Annexin-V and propidium iodide (PI) double staining confirmed that 18b induces apoptosis in cytotoxic HCT-116 cells. Moreover, the influence of 18b on cell cycle distribution was assessed on the HCT-116 cell line, and a cell cycle arrest was observed at the G1/S phases.
