Oxalyl chloride (130 mu; 1.49 mmol; 3 eq) and anhydrous dimethylformamide (catalytic amount) were added dropwise to a solution of 2-[l-(4-fluorophenyl)-3-[2- (4-methoxyphenyl)ethyl]-5-oxo-2-sulfanylideneimidazolidin-4-yl] acetic acid (1-21) (200 mg; 0.50 mmol; 1 eq) in anhydrous dichloromethane (8 mL). The reaction mixture was stirred at room temperature for 2 hours. 5-Methoxypyrimidin-2-amine (1-22) (124 mg; 0.99 mmol; 2 eq) and pyridine (93mu; 1.49 mmol; 3 eq) were added and the reaction mixture was stirred at room temperature for 2 hours and 30 minutes. Water (20 mL) and saturated ammonium chloride (15 mL) were added and the aqueous layer was extracted with ethyl acetate (3 x 40 mL). The combined organic layers were washed with saturated sodium chloride (30 mL), dried over sodium sulfate, filtered and concentrated to dryness. The crude was purified on silica gel using dichloromethane/ethyl acetate (90/10) as an eluent. After trituration in ethanol/diethyl ether, the title compound 2-[l-(4-fluorophenyl)-3-[2-(4- methoxyphenyl)ethyl]-5-oxo-2-sulfanylideneimidazolidin-4-yl]-N-(5- methoxypyrimidin-2-yl)acetamide was obtained in 7% yield (23 mg) as an orange solid. 1H-NMR (Acetone-d6): delta (ppm) 2.93 (m, 1H), 3.1 (m, 1H), 3.43 (m, 1H), 3.72 (s, 3H), 3.81 (m, 2H), 3.95 (s, 3H) , 4.27 (m, 1H), 4.65 (s, 1H), 6.85 (d, 2H), 7.25 (m, 4H), 7.45 (m, 2H), 8.36 (s, 2H) , 9.58 (s, 1H); MS (ESI+): m/z = 509.9 [M+H]+ .
